Magnetic manipulation of nanorods inside the nucleus of living cells

Strong magnetic fields (~1 Tesla) are innocuous to organisms, however if magnetic particles are present in the organism, they can apply forces able to perturb their microenvironment. Previously, it has been possible to move a superparamagnetic bead by a distance of <200 nm inside the nucleus of a living cell. However, the method can be applied only a single cell at a time and the displacement of the bead is not large enough to produce a significant change of the nuclear organization. Here, we manipulate magnetic nanorods with custom magnetic properties inside the nucleus of living cells producing >2 um displacements in the nucleus interior. We use a system that can be applied to deform the nucleus of several cells at the same time and, because the magnetic field is generated by magnets located several centimeters away from the sample, it can be applied to cells of an organism.
