Vascular permeability changes in correlation with piggyBac transposon-based gene delivery rates

Abstract:

The mechanism of ultrasound-induced DNA delivery has been attributed mainly to the formation of pores on the surface of cell membranes. This pore formation mechanism should result in a decrease in the cell resistivity accompanied by a simultaneous increase in the cell permeability. However, previous research efforts have yet to link cell behavior in terms of real-time permeability quantification to the rate of the achieved DNA delivery. In this study, we correlate ultrasound-induced DNA delivery assisted by ultrasound contrast agents (UCAs) with real-time measurement of cell permeability variations. Helper-independent plasmids with the mouse codon-based piggyBac transposon (mPB) gene in combination with cationic ultrasound contrast agents (UCAs) are investigated for a range of acoustic parameters.
