Nanoneedle Probes for Minimally Invasive Cellular Delivery

Abstract:

Current microinjection delivery technology relies on the use of a micropipette with a relatively large size (~1 micron in tip size) and a tapered tip shape which often causes cell damage, especially for delivery to the nucleus or small cells. This damage can introduce error into experiments and increase the time and cost of experimentation. We have developed a nanoneedle probe delivery method which uses a nanoneedle that has a uniform diameter of 100 nm or less over a long length of 10-20 microns from the tip and is conductive, to serve as a carrier for delivering quantum dots (QDs) into cells. Nanoneedle probes can be used for minimally invasive delivery of distinct amounts of cargo into cells and nuclei. The probes can also be used as sensors to take measurements within the living cell as it is exposed to different conditions and materials. Finally a probe of this nature can also be used to carry-out “cellular surgery,” to disable certain organelles within the cell. These types of experiments will lead to a better understanding of cellular function and contribute to the development of new medicines.
