Label-free Raman Micro-Spectral Imaging of the Microenvironment of PANC-1 Spheroids

Abstract:

Spheroids are 3D tumor models, which reflect diverse properties of the growing tumor. Due to its avascular microenvironment, initial tumor growth patterns as well as subsequent tumor-core biochemistry may be studied in depth, from which insight into tumor mass differentiation and organization may be deduced. A label-free and non-destructive imaging method recently gaining recognition is Raman micro-spectroscopy. We have thus utilized the instrumentation in synergy with mathematical data-unmixing algorithms, which paint a biochemical picture of the spheroid milieu. This non-invasive method of analysis will ultimately give rise to a more in-depth comprehension of the biochemical organization of the spheroid, render a better understanding of tumor growth and development, elucidate drug penetration patterns, and subsequently offer insight into its suppression.
