Optimization of preparation conditions for size-controlled solid lipid nanoparticles

Abstract:

This study is to prepare solid lipid nanoparticles, with target sizes at 100, 500 and 1000 nm. Nanocrystallization method was applied to prepare solid lipid nanoparticles using beef tallow, hydrogenated soybean oil and hydrogenated palm oil which transit from liquid to solid at above 60 ℃. The factors affecting the size and the stability of lipid nanoparticle, such as dispersing method, cooling rate, lipid content and surfactant type and ratio were investigated to control the size of solid lipid nanoparticles. Size distribution, scanning electron microscopy, specific surface area and zeta-potential were measured to characterize the solid lipid nanoparticles. The size of solid lipid nanoparticles decreased with increasing the content of lipid material with low melting point, decreased with increasing lipid content and increased with increasing surfactant content. It indicates that the size of solid lipid nanoparticle can be controlled by adjusting the parameters during solid lipid nanoparticle formation. From above results, size-controlled solid lipid nanoparticle can be applied to estimate in vitro and in vivo
