A sensitive protein detection based on fluorescent dye-doped silica nanoparticles and lateral flow test strip

In this article, we report a lateral flow based biosensor for rapid and sensitive detection of Influenza A antigen using fluorescent dye-doped silica nanoparticles as labels. Combining high signal brightness and photo stability of fluorescent dye-dope silica nanoparticles with advantages in simplicity and rapidity of lateral flow assay, highly sensitive detection of protein is realized. The nucleoprotein (NP) which is one of the most abundant structural proteins present in influenza A virion is used as a model to demonstrate a good performance of this proposed biosensor. Under optimal conditions, the fluorescent biosensor is able to detect NP in nanogram (ng) range using a portable strip reader in 30 minutes without cross-reactivity with other proteins. The detection of NP in allantoic fluid is also performed to demonstrate the detection in biological samples.
