Comparison of Nanoporation of Different Cell Lines

The threshold of nanoporation was compared at various electric pulse durations for different cell lines: human erythrocytes, Chinese hamster ovary cells, and rat glioma C6 cells. Electroporated cells were determined from the release of intracellular potassium ions. The dependences of the fraction of electroporated cells on the pulse intensity were obtained for the cells exposed to single square-wave electric pulses with the durations varied from 20 us to 2 ms. Electric field strength required to electroporate the cells was dependent on the pulse duration, decreasing with increasing pulse duration. In adition, for human erythrocytes it was about two times higher than for the rest cells studied. Then, the average cell radii were measured for each cell line and the transmembrane potential induced by the external electric field was calculated. The obtained values of the transmembrane potential were in the range of 450–910 mV and decreased with increasing pulse duration. The obtained dependences of the transmembrane potential required to electroporate 50 % of cells on the pulse duration were close to each other for all cell lines studied.
