Nanopatterned Substrate for Controlling Cell Alignment and Cell Internal Organization

Interaction of cells with their extracellular microenvironments is critical in tissue dynamics. It has been reported by many that substrate micro- and nano-groove topography can induce cell alignment along the groove direction, a phenomenon called “contact guidance”. The effects of groove geometries on cellular orientation, adhesion, morphology and proliferation for different cell types and substrate materials were reported. However, there is no study on the effect of “contact guidance” on cellular internal organization. Here we report our observation of “contact guidance” of SiHa cell on oxygen plasma treated PDMS nanograting substrate, as well as the alignment of cell division axis by the nanograting substrate. Combined with the recent works showing that cell division axis and internal organization are governed not by the cell shape, but by the anisotropy of cortical cues, our observation could help us understand better the “contact guidance” phenomenon at nanoscale, which may have an impact on implantable nanomaterials.
