Mediators of Intracellular Nanoparticle Trafficking Enhance Transgene Expression Efficacy of Polymer-DNA Complexes

Abstract:

Sequestration of nanoscale cargo (27 nm quantum dots and 150-250 nm polymer-DNA ‘polyplexes’) at a single location near the microtubule organizing compartment (MTOC) correlated with low polymer-mediated transgene expression compared to distributed localization throughout the cytoplasm. Treatment with histone deacetylase inhibitors, (e.g. tubacin) which acetylate tubulin of microtubules in the cytoplasm, abolished quantum dot and polyplex sequestration at the MTOC and enhanced transgene expression. Mediators of microtubule-mediated intracellular trafficking are attractive for enhancing the efficacy of nanoscale therapeutics.
